MAGEC2 encodes melanoma-associated antigen C2 (MAGE-C2), a cancer-testis antigen with restricted expression in normal germline tissues and frequent re-expression in diverse tumors. MAGE-C2 participates in protein homeostasis by engaging ubiquitin-dependent processes, including modulation of E3 ligase activity through interactions with RING-domain partners, thereby influencing stability of signaling and cell-cycle regulators. Dysregulated MAGE-C2 expression has been linked to altered proliferation, stress responses, and transcriptional programs associated with malignant progression and immune recognition. As a tumor-associated antigen, it is widely studied in antigen presentation contexts and as a molecular correlate of tumor phenotype and pathway rewiring.
MAGE-C2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the MAGEC2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the MAGEC2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the MAGEC2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish MAGE-C2 protein expression.
This CRISPR knockout system enables efficient generation of MAGEC2-deficient cell models for investigation of MAGE-C2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
