ZBTB7A encodes the transcription factor LRF/Pokemon (also known as FBI-1), a BTB/POZ and zinc-finger protein that binds GC-rich DNA elements to modulate gene expression programs controlling cell fate decisions. It participates in chromatin-dependent transcriptional repression and interfaces with pathways governing proliferation, differentiation, and lineage commitment, including regulation of metabolic and stress-response genes in a context-dependent manner. LRF/Pokemon is linked to hematopoiesis and immune cell development and has been studied for its effects on erythroid maturation and lymphoid differentiation. Altered ZBTB7A activity or expression has been associated with dysregulated transcriptional networks in cancer and other proliferative disorders, supporting its relevance for mechanistic studies of oncogenic and developmental pathways.
LRF/Pokemon CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ZBTB7A gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ZBTB7A together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ZBTB7A open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish LRF/Pokemon protein expression.
This CRISPR knockout system enables efficient generation of ZBTB7A-deficient cell models for investigation of LRF/Pokemon signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
