ErbB-4 Antibody (D-10) is a mouse monoclonal IgG2b antibody that detects ErbB-4 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ErbB-4, a member of the epidermal growth factor receptor family, plays a crucial role in cell signaling pathways that regulate cell proliferation and differentiation, making ErbB-4 a significant target in cancer research, particularly in breast carcinoma. ErbB-4 is primarily located on the cell membrane, where ErbB-4 can form homodimers or heterodimers upon ligand binding, which is essential for activation and subsequent signaling cascades. Notably, the gene encoding ErbB-4 produces a full-length protein that is cleaved to generate a short membrane-anchored cytoplasmic domain fragment and a long ectodomain fragment; the short fragment is heavily phosphorylated on tyrosine residues and exhibits catalytic activity, further emphasizing the importance of structural integrity in mediating cellular responses. ErbB-4 expression is particularly high in breast carcinoma cell lines, as well as in normal tissues such as skeletal muscle, heart, pituitary, brain, and cerebellum, highlighting ErbB-4′s relevance in both normal physiology and pathological conditions.
