EMILIN2 encodes EMILIN-2, an extracellular matrix glycoprotein enriched in elastic fiber–associated microfibrils that contributes to matrix organization and cell–matrix signaling. EMILIN-2 interfaces with integrin-mediated adhesion and mechanotransduction, influencing cytoskeletal dynamics, migration, and tissue remodeling. Through these extracellular cues, EMILIN2 can modulate pathways linked to inflammation and vascular and stromal homeostasis, including processes that shape angiogenic and fibrotic responses. Dysregulated EMILIN2 expression and altered matrix architecture have been associated with tumor microenvironment remodeling and cardiovascular-related extracellular matrix defects, supporting its relevance in disease mechanism research.
EMILIN-2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the EMILIN2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the EMILIN2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the EMILIN2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish EMILIN-2 protein expression.
This CRISPR knockout system enables efficient generation of EMILIN2-deficient cell models for investigation of EMILIN-2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
