NCBP2 encodes CBP20, the small subunit of the nuclear cap-binding complex that recognizes the 7-methylguanosine cap on nascent RNA polymerase II transcripts. By coupling cap recognition to RNA processing, CBP20 supports pre-mRNA splicing, 3′ end formation, nuclear export, and RNA surveillance pathways including nonsense-mediated mRNA decay. Through these functions, NCBP2 helps coordinate gene expression programs and transcriptome integrity during cell growth and stress responses. Dysregulation of cap-dependent RNA metabolism and mRNA quality control has been linked to oncogenic transcriptional states and neurodevelopmental phenotypes, making NCBP2 a useful node for studying disease-associated RNA processing mechanisms.
CBP20 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the NCBP2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the NCBP2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the NCBP2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish CBP20 protein expression.
This CRISPR knockout system enables efficient generation of NCBP2-deficient cell models for investigation of CBP20 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
