    Overview

    CSNK2A2 encodes the human casein kinase II alpha′ catalytic subunit, a constitutively active serine/threonine kinase that forms the CK2 holoenzyme with regulatory beta subunits and phosphorylates a broad spectrum of nuclear and cytoplasmic substrates. CK2 signaling interfaces with core processes including cell-cycle progression, DNA damage responses, chromatin regulation, and apoptosis, and it modulates major pathways such as NF-κB, Wnt/β-catenin, PI3K/AKT, and MAPK through substrate phosphorylation. By shaping phosphorylation-dependent control of transcription factors and RNA-processing proteins, CSNK2A2 influences proliferation and stress adaptation. Dysregulated CK2 activity and altered CSNK2A2 expression have been associated with tumor biology, inflammatory signaling, and other phosphorylation-driven pathologies, supporting its utility as a mechanistic node in biomedical research.

    casein kinase IIα′ CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the CSNK2A2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the CSNK2A2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the CSNK2A2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish casein kinase IIα′ protein expression.

    This CRISPR knockout system enables efficient generation of CSNK2A2-deficient cell models for investigation of casein kinase IIα′ signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
