Agrin Antibody (D-2) is a mouse monoclonal IgM antibody that detects Agrin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Agrin antibody (D-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Agrin plays a crucial role in the neuromuscular junction by facilitating the clustering of acetylcholine receptors (AChRs) on the muscle cell surface, which is essential for effective synaptic transmission and muscle contraction. This clustering is vital for the formation and maintenance of the neuromuscular junction, ensuring that signals from motor neurons are efficiently transmitted to muscle fibers. Agrin is primarily located in the basal lamina of muscle cells, where Agrin interacts with other proteins to orchestrate the assembly of the postsynaptic apparatus. Notably, Agrin exists in several splice variants, with specific forms exhibiting a high affinity for AChRs, which underscores the importance of post-translational modifications in regulating Agrin function. Additionally, the interaction between Agrin and the receptor tyrosine kinase MuSK is critical for the signaling pathways that lead to AChR aggregation, highlighting the intricate molecular mechanisms that govern neuromuscular junction formation and stability.

The epitope is somewhere within the LG3 domain of agrin, is this specific to the agrin LG3 domain? (there are a number of other proteins/ proteoglycans containing this domain)

I used m-IgGκ BP-HRP: sc-516102 for agrin WB detection in human EC cell line HUVEC and mouse primary lung EC cells. I found one really week band at 95-130 kDa. Is it the right band?

Can sc-374117 antibody recognize the 22-kDa fragment of agrin (CAF) by western blotting?

Thank you for your question. Agrin Antibody (D-2): sc-374117 has been raised against amino acids 1913-1942, which are present in the Agrin C-terminal 22 kDa fragment that corresponds to amino acids 1864-2068. The antibody is also specific against this fragment.

Can anti-mouse IgG secondary antibody be used to detect this primary antibody in WB?

We would recommend using our m-IgGκ BP-HRP: sc-516102 for detection of this mouse IgM primary antibody in Western blot. Our mouse IgG kappa binding protein (m-IgGκ BP) conjugated to Horseradish Peroxidase (HRP) is confirmed to detect mouse IgG and mouse IgM (kappa light chain type).

Does this work on frozen mouse and frozen human tissue? For IHC and IF

I use this antibody at a 1:200 dilution often for IF work (paraffin sections and fixed cultured cells) in mouse, rat and human tissue and it is absolutely stunning.
