ERK Inhibitor II, FR180204 ist ein neuartiger ERK-selektiver Inhibitor, der die Zelle durchdringen kann. ERK Inhibitor II, FR180204 hemmt ERK1 (Ki=0,31 muM), ERK2 (Ki=0,14 muM), TGFbeta-induzierte AP-1-Aktivierung und wirkt als kompetitiver ATP-Inhibitor. Darüber hinaus kann es die antigenspezifische Aktivierung von T-Zellen unterdrücken. In Studien zur kollageninduzierten Arthritis wurde beschrieben, dass in Anwesenheit des ERK-Inhibitors II die FR180204-Hypersensibilität vom verzögerten Typ abgeschwächt wird und die Plasma-Antikörperspiegel gegen CII sinken. Studien zum Inhibitor-ERK2-Komplex zeigen, dass der ERK-Inhibitor II, FR180204 wenig Aktivität gegenüber IKKalpha, MEK1, MKK4, PDGFRalpha, PKCalpha, Src und Syk zeigt. ERK-Inhibitor II, FR180204 ist ein Inhibitor von p38 alpha.
