Varicella Zoster Virus IE62 Inhibitors

Varicella Zoster Virus IE62 inhibitors operate through multiple mechanistic avenues to downregulate or completely halt the function of the IE62 protein, which is essential for viral replication and immune evasion. Direct-acting inhibitors such as Acyclovir, Foscarnet, and Brivudine work by inhibiting viral DNA polymerase or thymidine kinase, leading to decreased transcription and translation of the IE62 gene. Bortezomib operates through the ubiquitin-proteasome pathway, affecting IE62 indirectly by the degradation of IκB and subsequent NF-κB activation. Quercetin and SP600125 are potent inhibitors of the PI3K/AKT and JNK signaling pathways, respectively. On the other hand, Mycophenolic Acid disrupts purine synthesis by inhibiting inosine monophosphate dehydrogenase, causing a decrease in both viral RNA and DNA synthesis, including that of IE62. Curcumin, Rapamycin, Roscovitine, and Axitinib work through unique pathways. Curcumin inhibits NF-κB, Rapamycin inhibits mTOR signaling, Roscovitine targets CDKs, and Axitinib inhibits VEGFRs, each contributing to the disruption of IE62's multifaceted roles in the viral life cycle and pathogenesis.
