USP27X Inhibitors

Chemical inhibitors of USP27X function by various mechanisms, each related to the modulation of ubiquitin levels within the cell. P22077 and HBX 41108 are selective inhibitors that target USP7, a deubiquitinating enzyme closely related to USP27X. By inhibiting USP7, these chemicals lead to an accumulation of ubiquitinated proteins, which can saturate the enzymatic activity of USP27X. The excess of ubiquitinated substrates competes for attention from USP27X, thereby limiting its ability to process additional proteins. Similarly, WP1130 and b-AP15 target different deubiquitinases, culminating in an increased ubiquitinated protein pool. WP1130 degrades multiple deubiquitinases, while b-AP15 specifically inhibits the proteasomal deubiquitinating activity. These actions lead to a surplus of ubiquitin-conjugated proteins that can overwhelm USP27X, reducing its functional capacity.

Other chemicals such as ML364, LDN-57444, and IU1 inhibit other specific deubiquitinating enzymes like USP2, UCHL1, and USP14, respectively. By blocking these enzymes, they indirectly contribute to the heightened ubiquitinated protein levels. This widespread increase in ubiquitin-protein conjugates indirectly impedes USP27X by creating a competitive substrate environment, thus diminishing its deubiquitinating efficiency. Furthermore, NSC632839 and PR-619 exhibit broad-spectrum inhibition of deubiquitinases, which also results in the accumulation of ubiquitinated proteins. This broad inhibition feeds into the substrate competition, effectively limiting USP27X's capacity to act on its specific substrates. On the flip side, chemicals like PYR-41 and RA-9 take a different approach by reducing ubiquitin availability or inhibiting its cleavage from substrates, thereby affecting the substrate turnover and indirectly limiting USP27X activity. Lastly, G5, as a selective inhibitor of the USP1/UAF1 complex, contributes to the pool of ubiquitinated proteins, further restricting the functional reach of USP27X due to competitive substrate binding.
