RNF151 Inhibitors

MLN4924 disrupts the neddylation process that is essential for the activation of cullin-RING ligases, a family that includes RNF151. PYR-41 and HBX 41,108 target early steps in the ubiquitination cascade by inhibiting the ubiquitin-activating enzyme E1 and the ubiquitin-conjugating enzyme E2, respectively. Their action reduces the ubiquitin moiety's availability for conjugation to substrate proteins, thereby affecting the ubiquitination process where RNF151 is a participant.

Proteasome inhibitors like MG132, Bortezomib, Lactacystin, Clasto-Lactacystin β-lactone, and Epoxomicin function by preventing the degradation of ubiquitinated proteins. This inhibition leads to an accumulation of ubiquitinated substrates, which can saturate the UPS and influence the regulatory role of RNF151 within this pathway. By increasing the levels of ubiquitinated proteins, these inhibitors can indirectly affect the selection and processing of substrates that are targeted by RNF151. IU1 and VLX1570 modulate the activity of deubiquitinases (DUBs), enzymes that remove ubiquitin from proteins, thus affecting the recycling of ubiquitin and potentially the activity of ubiquitin ligases like RNF151. By altering the balance of ubiquitination and deubiquitination, these inhibitors can lead to changes in the steady-state levels of proteins within the cell, affecting processes where RNF151 is involved. Lastly, NSC697923 and Thalidomide have been shown to interact with the UPS. NSC697923 affects the ubiquitin-conjugating enzymes UBE2N and UBE2V1, which can indirectly modulate the activity of RNF151.
