Chemical inhibitors of I-Plastin can functionally modulate its activity by targeting the actin cytoskeleton, to which I-Plastin binds. Phalloidin, for example, binds to F-actin, promoting filament stability and thereby preventing the depolymerization that is necessary for I-Plastin to exert its actin-bundling function. Cytochalasin D caps the barbed ends of actin filaments, hindering their elongation and thus indirectly inhibiting I-Plastin's ability to bind and stabilize these structures. Similarly, Latrunculin A binds to G-actin, sequestering it so that the polymerization of actin filaments is blocked, leaving I-Plastin without its substrate. Swinholide A severs actin filaments and hampers their reassembly, diminishing the actin structures required for I-Plastin activity. Jasplakinolide promotes actin polymerization and stabilizes filaments, which can saturate the binding sites for I-Plastin on actin, effectively inhibiting its bundling activity.
Misakinolide A also stabilizes and promotes the formation of actin bundles, which could compete with I-Plastin for binding sites, inhibiting its normal function. Tectochrysin and Wortmannin inhibit the PI3K/Akt pathway and phosphoinositide 3-kinases respectively, both of which are involved in actin cytoskeleton regulation and, therefore, can indirectly inhibit I-Plastin's interaction with the cytoskeleton. ML-7 and Y-27632 target myosin light chain kinase and Rho-associated protein kinase, both of which are implicated in actin-myosin interactions and actin filament organization-processes that are critical for I-Plastin's role in actin bundling. Lastly, CK-636 inhibits the ARP2/3 complex, which is essential for actin nucleation. By impeding the formation of new actin filaments, CK-636 indirectly limits the availability of actin polymers for I-Plastin, thereby inhibiting its function. Each of these inhibitors exerts an influence on the actin cytoskeleton that could lead to the functional inhibition of I-Plastin by preventing it from effectively interacting with or modifying actin filaments.
