Geminin Coiled-Coil Domain Containing (GMNC) activators are a class of chemicals that are postulated to enhance the functional activity of GMNC through various signaling mechanisms. Forskolin, by stimulating adenylate cyclase, raises intracellular cAMP levels, which in turn activates PKA. The activated PKA can phosphorylate a range of proteins, potentially including GMNC. This phosphorylation can regulate GMNC's activity in the context of its role in cell cycle regulation. IBMX works synergistically with Forskolin by inhibiting the degradation of cAMP, thus maintaining an environment conducive to PKA activation and subsequent phosphorylation effects on GMNC. Similarly, Isoproterenol functions as a β-adrenergic agonist, and Dibutyryl-cAMP acts as a membrane-permeable cAMP analog, both leading to PKA activation and potentially enhancing GMNC activity.

On the other hand, Phorbol 12-myristate 13-acetate (PMA) activates PKC, which is known to have a broad range of effects, including the modulation of cell cycle regulators, which could indirectly affect GMNC activity. Ionomycin, by raising intracellular calcium levels, can activate calcium-dependent signaling pathways that potentially interact with GMNC.
