GAMT Inhibitors

GAMT inhibitors encompass chemicals that modulate the activity or function of GAMT, a pivotal enzyme in creatine synthesis. Direct inhibitors are not frequently cited, but numerous compounds influence GAMT by mediating the pathways linked with its substrate or product.

S-adenosylhomocysteine (SAH) and guanidinopropionic acid act as direct competitors to the GAMT substrates. While SAH competes for the methyl donor SAMe, guanidinopropionic acid poses as a substrate analog, leading to reduced efficacy of GAMT. L-Arginine and SAMe, pivotal components in the creatine synthesis pathway, can exert indirect inhibitory effects on GAMT when present in high concentrations, primarily due to feedback mechanisms. Compounds like cyclocreatine, beta-guanidinopropionic acid, and creatinine have strong connections to the creatine metabolic pathway. Their presence and the changes they induce can influence GAMT activity. Nitric oxide, an influential molecule in various physiological processes, impacts the conversion of L-arginine to guanidinoacetate, and hence, can affect GAMT indirectly. Similarly, adenosine's role in various metabolic pathways can influence SAMe levels and thus, the activity of GAMT. Lastly, excessive glycocyamine (another term for guanidinoacetate) can exert feedback control on GAMT, inhibiting its function indirectly.
