FBXO21 Inhibitors

Chemical inhibitors of FBXO21 function through different mechanisms to impede the protein's role in the ubiquitination process. MLN4924 targets the NEDD8-activating enzyme, essential for the neddylation that activates the SCF complex, where FBXO21 is a component, thus indirectly inhibiting FBXO21's ability to tag proteins for degradation. Similarly, compounds such as Pyr41 and IX 207-887 inhibit the E1 ubiquitin-activating enzyme, which is crucial for the initiation of the ubiquitination cascade. By halting this initial step, these inhibitors prevent FBXO21 from tagging substrates with ubiquitin. TAK-243 also inhibits the E1 enzyme, leading to a blockade of ubiquitin activation and subsequent interference with the ubiquitination process involving FBXO21.

On the other hand, several inhibitors focus on the proteasome to indirectly affect FBXO21 activity. Compounds such as MG132, Lactacystin, Bortezomib (also known as Velcade), Epoxomicin, Carfilzomib, and Oprozomib prevent the degradation of polyubiquitinated proteins by inhibiting the proteasome. This inhibition results in the accumulation of FBXO21 substrates within the cell. The increased levels of these substrates can lead to an indirect suppression of FBXO21's function as part of the cell's regulatory feedback mechanisms. By blocking the final step in the ubiquitin-proteasome pathway, these inhibitors ensure that proteins marked by FBXO21 are not degraded, thereby influencing the activity of FBXO21 by the buildup of its intended targets. Clasto-Lactacystin β-lactone, the active form of Lactacystin, similarly binds to the proteasomal subunit and irreversibly inhibits its activity, leading to the same effect on the feedback regulation of FBXO21.
