ENT2 Inhibitors

The Equilibrative Nucleoside Transporter 2 (ENT2) is a membrane protein primarily involved in the translocation of purine and pyrimidine nucleosides across cell membranes. This transporter plays a crucial role in regulating nucleoside homeostasis within cells, thereby influencing various cellular processes such as nucleic acid synthesis, energy metabolism, and signaling pathways. ENT2 facilitates the bidirectional transport of nucleosides, allowing cells to uptake extracellular nucleosides for intracellular utilization and to export intracellular nucleosides into the extracellular environment. Inhibition of ENT2 can be achieved through several mechanisms, predominantly through the competitive binding of inhibitors at the nucleoside-binding site of the transporter. Competitive inhibitors structurally resemble nucleosides and effectively compete with them for binding to the active site of ENT2, thereby reducing the rate of nucleoside transport. Additionally, non-competitive inhibition may occur through the binding of inhibitors at allosteric sites, altering the conformational dynamics of the transporter and impairing its function. Furthermore, substrate depletion strategies can indirectly inhibit ENT2 activity by reducing the availability of nucleoside substrates for transport. These inhibition mechanisms collectively disrupt nucleoside transport mediated by ENT2, leading to perturbations in nucleoside homeostasis and consequent alterations in cellular processes reliant on nucleoside flux. Understanding the intricate regulation of ENT2 and its susceptibility to inhibition offers insights into strategies for modulating nucleoside transport in physiological and pathological contexts.

Bosentan is a dual endothelin receptor antagonist that targets both endothelin receptor type A (ENTA) and endothelin receptor type B (ENTB). It was investigated for pulmonary arterial hypertension (PAH). By blocking the action of endothelin, it helps dilate blood vessels and reduce pulmonary hypertension.
