CYP11A1 Activators

CYP11A1 activators represent a diverse collection of chemicals that share the capacity to stimulate expression of the cytochrome P450 enzyme CYP11A1. As a class, these activators span different chemical types including steroids, industrial compounds, and environmental contaminants. A common attribute of many CYP11A1 activators is their ability to engage nuclear receptors that regulate CYP11A1 gene transcription. Binding of an activator to receptors like the glucocorticoid receptor, estrogen receptor, pregnane X receptor, constitutive androstane receptor, or peroxisome proliferator-activated receptors initiates intracellular signaling cascades that mobilize transcription factors and transcriptional co-regulators. These proteins then interact with hormone response elements in regulatory sequences of the CYP11A1 gene to enhance recruitment of RNA polymerase and elevate the production of CYP11A1 mRNA transcripts.

Alternatively, some CYP11A1 activators are thought to wield indirect effects through disruption of cellular processes regulating gene expression.They may perturb signaling networks, epigenetic machinery, or basic homeostatic pathways in a manner that secondarily modulates receptor signaling cross-talk or the function of DNA binding proteins governing CYP11A1 transcriptional controls. Through diverse direct and indirect mechanisms functioning at the level of gene regulation, this motley collection of chemicals collectively serves to boost CYP11A1 expression levels in target tissues where it is found.
