CRTAM, or Class I-restricted T cell-associated molecule, is a cellular adhesion molecule that belongs to the immunoglobulin superfamily. It is selectively expressed on the surface of CD4+ and CD8+ T cells upon their activation, as well as on natural killer (NK) cells. CRTAM is involved in the immune system's adaptive arm, playing a critical role in the modulation of immune responses.
The expression of CRTAM is transient, typically appearing within 24 hours after T cell activation and disappearing after 48 hours. This transient expression suggests a role in the early stages of immune cell response. CRTAM binds to its ligand, nectin-like 2 (Necl-2), which is expressed by dendritic cells and epithelial cells. The interaction between CRTAM and Necl-2 is important for the establishment of the immunological synapse, a specialized junction between T cells and antigen-presenting cells (APCs) that facilitates effective cell-cell communication and signaling.The engagement of CRTAM with its ligand during T cell-APC interaction is thought to contribute to the polarization of T cell responses. It is implicated in promoting the cytotoxic function of T cells and the production of cytokines like IFN-γ, which are crucial for the clearance of pathogens and tumor surveillance.
