ASB-7 Inhibitors

ASB-7 inhibitors act primarily through the ubiquitin-proteasome system, given that ASB-7 is an E3 ubiquitin ligase. Its main function is to mark proteins for degradation via the proteasome. Bortezomib, MG-132, Epoxomicin, and Lactacystin are proteasome inhibitors. They block the proteasome and thereby prevent the degradation of proteins ubiquitinated for destruction. ASB-7's function is indirectly inhibited due to the accumulation of proteins it has marked for destruction.

PYR-41 is an inhibitor of the ubiquitin-activating enzyme E1, which initiates ubiquitination. Without this initial step, ASB-7, being an E3 ubiquitin ligase, cannot mark proteins for degradation, leading to decreased functional activity. Similarly, MLN4924 inhibits the NEDD8 pathway involved in the modification of cullin proteins, which are an essential part of the Cullin-RING E3 ubiquitin ligases that ASB-7 belongs to. The functional activity of ASB-7 is disrupted by inhibiting the NEDD8 pathway. PR-619, a deubiquitinase inhibitor, disrupts the balance between ubiquitination and deubiquitination, which can inhibit ASB-7's function.
