Chemical activators of protein phosphatase 1, regulatory subunit 35 can modulate the protein's activity through various biochemical mechanisms. Forskolin acts directly on adenylate cyclase to increase levels of cyclic AMP within the cell, which subsequently activates protein kinase A (PKA). PKA then phosphorylates several substrates, including protein phosphatase 1, regulatory subunit 35, altering its conformation and interaction dynamics, thereby increasing its activity. Cyclic AMP follows a similar pathway, serving as a messenger to activate PKA and promote the phosphorylation and activation of protein phosphatase 1, regulatory subunit 35. Spermine, through its interactions with ion channels and modulation of kinase activity, also facilitates the activation of the protein by enhancing kinase-mediated phosphorylation.

On the other hand, several inhibitors of protein phosphatases, such as Calyculin A, Okadaic Acid, Tautomycin, Cantharidin, Microcystin-LR, Endothall, Fostriecin, and Rubratoxin B, indirectly contribute to the activation of protein phosphatase 1, regulatory subunit 35. These compounds inhibit the action of protein phosphatases 1 and 2A, leading to reduced phosphatase activity in the cell. The decreased phosphatase action results in an accumulation of phosphorylated proteins, which may include protein phosphatase 1, regulatory subunit 35. The cell responds to these changes by activating compensatory mechanisms to restore the balance of phosphorylation, leading to the activation of protein phosphatase 1, regulatory subunit 35. Zoledronic Acid, although primarily known for its inhibition of farnesyl pyrophosphate synthase, influences signaling pathways that can result in the increased activity of protein kinase A, which, in turn, can phosphorylate and activate protein phosphatase 1, regulatory subunit 35.
