I have a double signal from vinculin, one between 150-100 that corresponds to the information on the data sheet, but another very intense band between 75-100. Could someone tell me why?. It is mouse brown adipose tissue and it is mouse vinculin.

Hello, I'm a graduate student at Ryerson University, Toronto, Canada. I am planning to do immunofluorescence, and I was wondering if it's ok to fix with 4% PFA (50min), neutralize with 1% glycine and permeabilize with -20 degree C methanol. Thank you!

Can vinculin Antibody (7F9): sc-73614 be used in IHC with FFPE?

Yes, vinculin Antibody (7F9): sc-73614 is validated for use in IHC with formalin fixed, paraffin-embedded tissue sections. Rated 5 out of 5 by from The sc-73614 ab works very wellI prepared 10µg of whole cell lysates of HuH7 or PLC cells and run them on a 10% SDS-PAGE gel. Proteins were transferred from the gel into a nitrocellulose membrane and after blocking, membranes were incubated with the respective antibody rocking them in a shaker at 4 degree over night. The membranes were then washed and incubated with the appropriate secondary antibody and developed with the LI-COR Odyssey imager. The antibody against Vinculin specifically recognized one single protein and the staining produced a band at the expected molecular weight.

Cells were fixed with 0.5% buffered paraformaldehyde (PFA) for 10 minutes, and immunostained with vinculin antibody (sc- 73614, red). Cells were stained with Alexa 488-conjugated phalloidin to reveal actin filament organization (green) and with Dapi to label nuclei (blue).
