Mouse Tsn encodes translin, a conserved RNA- and DNA-binding protein that forms a heteromeric complex with trax (TSNAX) and participates in post-transcriptional gene regulation. Translin has been linked to RNA transport and stability, microRNA processing via the C3PO nuclease complex, and modulation of localized translation in neuronal and germ cell contexts. Through these activities, TSN influences stress-responsive signaling and gene expression programs that shape cell proliferation and differentiation. Altered TSN-associated RNA regulatory networks have been implicated in neurobehavioral phenotypes and metabolic regulation, making Tsn a useful locus for dissecting RNA-mediated mechanisms in disease-relevant models.
translin CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Tsn gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Tsn together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Tsn open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish translin protein expression.
This CRISPR knockout system enables efficient generation of Tsn-deficient cell models for investigation of translin signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
