Sarcospan Antibody (E-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects Sarcospan protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Sarcospan antibody (E-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Sarcospan, also known as K-Ras oncogene-associated protein, plays a crucial role in the dystrophin-glycoprotein complex (DGC), which is essential for maintaining muscle integrity and function. Sarcospan is primarily located in the sarcolemma of striated muscle cells, where Sarcospan forms a vital link between the subsarcolemmal cytoskeleton and the extracellular matrix. Proper localization of Sarcospan is critical, as defects in DGC components can lead to muscular dystrophy, highlighting Sarcospan′s importance in muscle viability. Sarcospan contains four transmembrane spanning helices with intracellular N- and C-terminal domains, and Sarcospan expression is significantly reduced in muscle tissues from patients with Duchenne muscular dystrophy. Sarcospan mRNAs are expressed across various cell lines, tumors, and normal tissues, with particularly high levels in muscle. Two isoforms, SPN1 and SPN2, arise from alternative splicing, with SPN1 predominantly found in heart and skeletal muscle, while SPN2 is expressed in a broader range of tissues, including the thymus, prostate, testis, ovary, small intestine, colon, and spleen. The sarcoglycan complex, which is integrally associated with Sarcospan in the DGC, is a heterotetrameric unit that also interacts with the signaling protein neural nitric oxide synthase through α-Syntrophin, further emphasizing Sarcospan′s multifaceted role in muscle function and signaling pathways.
