Can this be used for flow cytometry? If so, is it surface or ICS?

Thank you for your question. Pim-1 Antibody (12H8) is available conjugated to Alexa Fluor® 546 or Alexa Fluor® 594 for flow cytometry. The antibody was raised against full length of Pim-1 of mouse origin. Since no epitope mapping has so far been performed, there is no information on whether it binds a cytoplasmic or extracellular domain of the protein.

could It recognize PIM1-L, The longer varient?

Thank you for your question. This mouse monoclonal antibody, Pim-1 (12H8): sc-13513, is cross reactive with both isoforms of Pim-1. This is based on several published articles which reference this clone detecting both (e.g., PMID 19345670).

I used at 1:1000 dilution and detected with ChemdocMP and Thermo Fisher Pico (w. 1/10 Femto) substrate. Show weak but specific bands of 3 isoforms. Can extend exposure to 600 - 1200 seconds. Background is low.

The lysis of mitochondria is required to reveal the 40 kDa isoform-3 of Pim-1. It also works for Immunofluorescence but little longer exposure time needed.
