    SEE ALSO...

PCLN-1 Antibody (7A2) is a mouse monoclonal IgM antibody that detects PCLN-1 of mouse, rat, and human origin by immunofluorescence (IF) and enzyme-linked immunosorbent assay (ELISA). Anti-PCLN-1 antibody (7A2) is available as the non-conjugated format. PCLN-1, also known as paracellin-1, plays a crucial role in forming and maintaining tight junctions, which regulate the paracellular pathway between epithelial and endothelial cells. These tight junctions create a barrier preventing free passage of solutes and ions, maintaining cellular homeostasis and tissue integrity. PCLN-1 localizes to kidney tissue tight junctions, forming intercellular pores vital for magnesium and calcium resorption in the thick ascending limb of Henle. PCLN-1′s structure includes four transmembrane domains and intracellular amino and carboxy termini, characteristic of the claudin family. Mutations in PCLN-1 can lead to renal magnesium wasting, contributing to a rare autosomal recessive disorder known as renal hypomagnesemia with hypercalciuria and nephrocalcinosis, highlighting PCLN-1′s significance in renal function and electrolyte balance.

What is the recommended fixation method for immunofluorescence staining with PCLN-1 (7A2): sc-130561 monoclonal antibody?

    Asked by: Germaine
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
