Nucleoredoxin Antibody (E-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects Nucleoredoxin (NXN) in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-Nucleoredoxin antibody (E-12) comes in non-conjugated form and various conjugates, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® options. Nucleoredoxin, also known as NRX or TRG-4, is a 435 amino acid protein found mainly in the cytoplasm and nucleus, functioning as a redox-dependent negative regulator of the Wnt signaling pathway. This regulation supports essential cellular processes, including cell proliferation, differentiation, and embryonic development. Nucleoredoxin shows widespread expression in adult tissues, with notable presence in the nervous system and limb buds during key embryonic stages (day 10.5-11.5). A conserved thioredoxin domain and catalytic motif enable oxidoreductase activity, suggesting involvement in transcriptional regulation and modulation of protein phosphatase 2A (PP2A-Cα) activity. During oxidative stress, Nucleoredoxin interacts with dishevelled 1 (Dvl) through PDZ domain binding, demonstrating significance in stress-related cellular signaling. Located on human chromosome 17p13.3 and mouse chromosome 11 B5, Nucleoredoxin maintains evolutionary conservation across species.
