Notch3 encodes the Notch 3 receptor, a single-pass transmembrane protein that mediates juxtacrine signaling to control cell fate decisions, proliferation, and differentiation in a context-dependent manner. Upon ligand engagement, receptor processing enables release of the Notch intracellular domain, which translocates to the nucleus and regulates transcriptional programs with CSL/RBPJ and co-activators. In mouse tissues, Notch3 is prominently linked to vascular smooth muscle/pericyte biology and contributes to vessel maturation, tissue homeostasis, and stress responses. Dysregulated Notch3 signaling has been associated with aberrant differentiation states and vascular pathology, making it a relevant node for studying Notch pathway crosstalk and lineage control.
Notch 3 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Notch3 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Notch3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Notch3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Notch 3 protein expression.
This CRISPR knockout system enables efficient generation of Notch3-deficient cell models for investigation of Notch 3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
