Does the concentration that is given on the agarose-coupled (AC) version mean that there is 500 µg of antibody/ml bead suspension or is it 500 µg Antibody-Bead conjugate/ml?

Thank you for your question. The agarose conjugated antibodies (AC) contain 500µg of antibody in 1ml, 25% of the total volume is made up of Agarose (250µl).

How is this antibody conjugated to agarose? Will it be eluted unter reducing conditions or remain attached so that the AB can be removed after an IP? Can you also please give me the MW of the heavy and light chain of this antibody, respectively. Thx

Thank you for your question. Our antibodies are covalently bound to the agarose beads via the primary amines. Yes, after the IP, the agarose and antibody complex will break apart when added to the SDS buffer for the WB. The heavy chain is about 50kDa and the light chain is about 25kDa.

Hi, since this antibody is raised against N-terminal extracellular domain, it should recognize the transmembrane NRG1 (around 120kDa) from cell lysates. Then why the bands from your western blot examples are all around 35 kDa? Is it a specific isoform?

Thank you for your question. The molecular weight of Neuregulin-1 isoforms range within 26-71kDa. The exact epitope for this antibody has not been epitope mapped, but it maps somewhere within amino acids 21-230 of human Neuregulin-1, accession#Q02297. Can Neuregulin-1 (E-12): sc-393006 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, Neuregulin-1 (E-12): sc-393006 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Detected strong bands for menbrane-bound (~120kDa) as well as secreted (~50kDa) NRG-1 from human RF24 cells and A549 cells. keep in mind the secreted NRG-1 can be presented at different sizes in different cell lines. in RF24 and A549 cells, the bands were around 50kDa
