Nanos3 encodes an evolutionarily conserved RNA-binding protein that helps maintain primordial germ cell survival and germline identity during mouse development. By associating with target mRNAs and modulating their stability and translational output, NANOS3 contributes to post-transcriptional gene regulation programs that coordinate germ cell migration, cell-cycle control, and suppression of inappropriate somatic differentiation. Disruption of Nanos3 is linked to germ cell depletion and infertility phenotypes, making it relevant to studies of reproductive biology and mechanisms that safeguard genome integrity in the germline. As a germline regulator, NANOS3 is also used as a marker and functional node for investigating developmental trajectories and stress responses in early gonadal lineages.
Nanos3 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Nanos3 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Nanos3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Nanos3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Nanos3 protein expression.
