LCN15 encodes lipocalin-15, a member of the lipocalin family of secreted proteins that bind and transport small hydrophobic ligands, including lipids and retinoid-like metabolites. Through ligand sequestration and delivery, lipocalin-15 is positioned to influence extracellular signaling, membrane homeostasis, and intercellular communication in tissue microenvironments. Altered expression of lipocalins has been associated with inflammation, metabolic stress responses, and epithelial remodeling, making LCN15 a useful target for dissecting lipid-handling and stress-adaptive pathways. Studying LCN15 function can inform mechanisms linking secreted carrier proteins to paracrine regulation and disease-associated changes in cellular state.
Lipocalin-15 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the LCN15 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the LCN15 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the LCN15 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Lipocalin-15 protein expression.
This CRISPR knockout system enables efficient generation of LCN15-deficient cell models for investigation of Lipocalin-15 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
