ITI-H4 Antibody (F-9) is a mouse monoclonal IgG2b kappa light chain antibody that detects ITI-H4 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-ITI-H4 antibody (F-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The inter-α-trypsin inhibitor (ITI) family, to which ITI-H4 belongs, consists of structurally related plasma serine protease inhibitors synthesized in the liver, comprising different combinations of three highly homologous heavy chains (ITI-HI, ITI-H2, and ITI-H3) and one light chain (Bikunin). Notably, ITI-H4 features a proline-rich region (PRR) at its C-terminus, which may play a role in functional properties. The ITI family is a glycoprotein composed of three polypeptides linked by chondroitin sulfate: two heavy chains, ITI-H1 and ITI-H2, and Bikunin, with the latter conferring the protease-inhibitor function of ITI. The heavy chains, known as serum-derived hyaluronan-associated proteins (SHAPs), bind covalently to hyaluronic acid, contributing to the stabilization of the pericellular matrix. Importantly, while the ITI family is primarily composed of multi-polypeptide molecules, ITI-H4 is unique as a single-chain molecule. The gene encoding human ITI-H4 is located on chromosome 3p21.1, and interestingly, the gene transcriptions and products for rat and human ITI-H4 exhibit significant differences, indicating potential species-specific functions for this protein.
