HMG20A encodes HMG-20A, a high mobility group–like chromatin-associated factor that contributes to transcriptional regulation by influencing nucleosome organization and access of regulatory complexes to DNA. It has been implicated in epigenetic control of lineage programs, including neurodevelopmental gene expression, through interactions with chromatin remodeling and histone-modifying machinery. By shaping promoter and enhancer activity, HMG-20A can affect cell-cycle control, differentiation, and maintenance of cell identity. Altered regulation of HMG20A-linked chromatin states is relevant to studies of neurological phenotypes and cancer-associated transcriptional dysregulation.
HMG-20A CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the HMG20A gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the HMG20A together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the HMG20A open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish HMG-20A protein expression.
This CRISPR knockout system enables efficient generation of HMG20A-deficient cell models for investigation of HMG-20A signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
