ErbB-3 Antibody (2Q320) is a mouse monoclonal IgG2b kappa light chain antibody designed to target the ErbB-3 protein by recognizing an epitope mapped within the cytoplasmic domain of human ErbB-3. ErbB-3 monoclonal antibody (2Q320) exhibits broad species reactivity, effectively detecting ErbB-3 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). ErbB-3, also called FERLK, HER3, LCCS2, MDA-BF-1, VSCN1, erbB3-S, and ERBB3, is a critical member of the ErbB family of receptor tyrosine kinases, and is predominantly located on the cell membrane where ErbB-3 plays a pivotal role in cellular signaling pathways that regulate cell growth, differentiation, and survival. Structurally, ErbB-3 consists of an extracellular ligand-binding domain, a single transmembrane region, and an intracellular domain rich in tyrosine residues essential for downstream signaling. Post-translational modifications, particularly phosphorylation of specific tyrosine residues within the cytoplasmic domain, are crucial for ErbB-3′s function. These phosphorylations create binding sites for signaling molecules such as the p85 subunit of phosphoinositide 3-kinase (PI3K), thereby activating the PI3K/Akt pathway, which is vital for promoting oncogenic processes including tumor growth and resistance to apoptosis. Additionally, ubiquitination of ErbB-3 can regulate stability and degradation, further influencing signaling intensity and duration. The precise regulation of these post-translational modifications is essential for maintaining normal cellular functions, and dysregulation is often implicated in various cancers, where ErbB-3 is frequently overexpressed or mutated, contributing to tumor progression and therapeutic resistance. Anti-ErbB-3 antibody (2Q320) is available in both non-conjugated form and multiple conjugated variants, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates.
