As the question above noted, what is the broad region of possible recognition by this antibody? (i.e. something like "AA1-200"). I plan to use this on a split-Cas9 and need to know whether it will be specific for the N-terminal piece.

Thank you for your question. This antibody was raised against a recombinant protein corresponding to the N-terminal region of Cas9 of Streptococcus pyogenes origin. The exact binding epitope has not been mapped. It recognizes Cas9 variants including dCas9 (nuclease deficient Cas9). If you have any further questions, please contact Technical Service.

Is there any validation of this antibody working for immunofluorescence applications?

Thank you for your question. Yes, this mouse monoclonal antibody, Cas9 (7A9-3A3): sc-517386, has been validated to work for immunofluorescent staining applications. Unfortunately, we do not yet have any published IF data for this clone on our website. However, this clone, (7A9-3A3) has been widely published in the literature for use in a variety of applications, including IF. We do plan to update the data on our website once we have completed testing of the antibody.

Rated 5 out of 5 by from Works for Western blotThe antibody is effective for SDS immunoblotting. To dilute the primary antibody, we used a ratio of 1:500 in a mixture of 5% nonfat dry milk, 1X TBS, and 0.1% Tween® 20. The antibody was left overnight at 4°C.
