CalciFluor™ Fluo-8, AM has been developed to improve cell loading and calcium response while maintaining the convenient Fluo-3 and Fluo-4 spectral wavelength of maximum excitation at ~490 nm and maximum emission at ~520 nm. Similarly to Fluo 4 AM, the anionic carboxy groups are protected with acetoxy methyl esters allowing membrane permeability, which are subsequently removed by intracellular esterases allowing Ca2+ detection. CalciFluor™ Fluo-8, AM is efficiently loaded into cells at room temperature, whereas Fluo-3 and Fluo-4 require much harsher loading conditions. It is also two times brighter than Fluo-4 and four times brighter than Fluo-3. These features, as well as a lower price, make CalciFluor™ Fluo-8, AM a superior in vivo calcium indicator.
