c-Mpl抗体(E-7)是一种IgM κ型小鼠单克隆c-Mpl抗体(也被称为c-Mpl抗体),能够通过蛋白质印迹(WB)、免疫沉淀(IP)、免疫荧光(IF)和酶联免疫吸附测定(ELISA)等方法检测小鼠、大鼠和人源的c-Mpl蛋白。c-Mpl抗体(E-7)可作为非偶联的抗c-Mpl抗体使用。血小板生成素(TPO或THPO),也被称为c-Mpl配体(c-Mpl L),是一种细胞因子,在巨核细胞生成中发挥核心作用,影响巨核细胞的发展和成熟以及造血干细胞产生的血小板生产。TPO通过TPO受体c-Mpl发挥其生物学效应。c-Mpl是细胞因子受体超家族的成员,其表达仅限于造血组织和细胞,如骨髓、脾脏、胎儿肝脏和CD34+细胞。用TPO刺激c-Mpl会激活Janus酪氨酸激酶家族成员Tyk 2和JAK2,进而磷酸化Stat5和Stat3,导致它们的核转位和Stat响应基因的转录。c-Mpl的突变已被认为是导致某些人类疾病的原因,包括先天性无巨核细胞性血小板减少症(CAMT)和伴无桡骨血小板减少症(TAR)综合征。

If the antibody to c-Mpl E-7 is a monoclonal mouse Ig M isotype, why do you suggest to use m-IgGκ BP-HRP as a secondary reagent? Is the secondary able to bind IgM as well?

Thank you for your question. IgGκ BP-HRP is the preferred secondary detection reagent for c-Mpl Antibody (E-7) for WB applications but, yes an IgM secondary would work as well.
