ACE 抗体

ACE2抗体（E-11）是一种IgG1 κ型小鼠单克隆ACE2抗体（也称为ACE-2抗体、ACEH抗体或血管紧张素转换酶2抗体），可通过WB、IP、IF、IHC（P）和ELISA检测小鼠、大鼠和人源ACE2蛋白。ACE2抗体（E-11）既有非偶联抗ACE2抗体形式，也有多种偶联形式的抗ACE2抗体，包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®偶联物。血管紧张素转换酶（ACE）是一种羧基末端二肽基外肽酶，可将血管紧张素I转化为强效的血管收缩激素--血管紧张素II。ACE有两种同工型，即肺ACEP和睾丸ACET。ACEP是一种在肺、肝、肾上腺皮质、胰腺、肾和脾的血管内皮细胞中表达的糖蛋白。ACET同工型仅在成年睾丸中由发育中的精子细胞特异性表达，特别是晚期粗线期精母细胞。此外，ACE可使血管舒张肽缓激肽失活，并参与血压调节和体液/电解质稳态。ACE2是已知的第一个ACE的人类同源物。与在血管系统中普遍表达的ACE不同，ACE2仅在心脏、肾脏和睾丸细胞中表达。

Will this product work for performing ICC on pig cell culture samples? (Porcine Valve Interstitial Cells)

Thank you for your question. We have only validated this antibody to work in mouse, rat and human. There is a 33% homology in porcine.

I just got a free sample of ace 2 (E 11) and I did western blot on mouse plasma samples with 1:500 and 1:100 dilution. I used m-IgGκ BP-HRP: sc-516102 as secondary antibody but the blot didnot give anything.

We typically can not provide troubleshooting support or replacements for the free sample antibodies as there is not enough of the antibody to perform repeated testing. Hi I performed WB use this antibody but unlike the people above, it doesn't working in un transfected HEK293T cells and A549 WCL. Is it right?. I saw the some peoples review they were detected band in non-transfected cells.

Thank you for your question. Based on our testing and the Western blot data shown on our website, we did not observe ACE2 in non-transfected HEK293T cells. According to the Human Protein Atlas, there is very low RNA expression in these cell lines. https://www.proteinatlas.org/ENSG00000130234-ACE2/cell

After that, cells were washed in FACS buffer (D-PBS/0.1% BSA), fixed in 4% PFA at room temperature (RT), and analyzed by flow cytometry.

I noted some nuclear staining but this was common to all other tested Ab and it might be related to VeroE6 cells. Attached image was collected at cell basal plane by confocal Zeiss 800. Immunofluorescence procedure: Storti et al., 10.1016/j.bpc.2019.106225.
