ACCα抗体(D-5)是一种IgG2b κ小鼠单克隆ACC alpha抗体(也称为ACACA抗体、乙酰辅酶A羧化酶Alpha抗体、乙酰辅酶A羧化酶1抗体、ACC1抗体、ACC-Alpha抗体、ACAC抗体或ACCA抗体),可通过WB、IP、IF和ELISA检测人源的ACC alpha蛋白。ACCα抗体(D-5)既有非偶联抗ACC alpha抗体形式,也有多种偶联形式的抗ACC alpha抗体,包括琼脂糖、HRP、PE、FITC和多种Alexa Fluor®偶联物。乙酰辅酶A羧化酶(ACC)是一个复杂的多功能酶系统,可催化乙酰辅酶A羧化成丙二酰辅酶A,这是脂肪酸合成中的限速步骤。运动可降低人体肌肉中乙酰辅酶A羧化酶的活性。ACCα(ACC1)是长链脂肪酸生物合成中的限速酶,而ACCβ(ACC2)可控制线粒体脂肪酸氧化。ACC的这两种异构体控制细胞中的脂肪酸含量。ACCα的催化功能受靶向丝氨酸残基的磷酸化(失活)和去磷酸化(激活)以及柠檬酸或棕榈酰辅酶A的别构转化的调节,这是该酶的短期调节机制。编码ACCα的基因定位于人类17号染色体,并编码一种ACC形式,这是脂肪生成组织中的主要ACC。ACCβ的催化核心与ACCα的同源,区别在于其在N端有一个额外的约150个氨基酸的肽段。

What blocking solution should I use with ACCα (D-5): sc-137104 monoclonal antibody for Western blot?

The antibody is specific for ACCα, but the signal is not very strong and the background is a little bit high. Application: Western blot Sample: whole lysate of human hepatocytes from Huh7 cell line Gel running conditions: reduced, denaturing, 4-12% SDS-PAGE Loading amount: 30 µg protein/lane Transfer: semi-dry, on nitrocellulose membrane (0.45 µm) Blocking agent: 5% milk in TBST buffer, for 1h, RT Primary antibody dilution- 1: 400 in 5% milk, for 16 h at 4°C Secondary antibody: non- Santa Cruz antibody anti-mouse-HRP, 1:10000 dilution in 5% milk, for 1h, RT Detection: ECL substrate (femto sensitivity) Exposure time: 20 sec Bands: specific, at 265 kDa; non-specific bands are absent.
