Thank you for your question. Yes, sc-81779: ABC1 (A00121.01) is recommended for use in IHC with paraffin-embedded sections. Rated 5 out of 5 by from specific staining with high dilutionThe primary antibody could be further diluted to decrease non specific staining. The tissue was homogenised in RIPA buffer containing 1mM PMSF, 1µM aprotinin, 1µg/ml Pepstatin A, 0.5 µg/ml okadaic acid, 1 mM benzamidine HCl and briefly sonicated. The supernatant was used as the total lysate. The sample was treated with Laemli buffer at 2 different conditions before loading. 1. lysate was incubated in Laemli buffer for 30 min at RT. 2. lysate was heated to 70 C for 10 min. Both yielded similar results. Proteins were transfered onto nitrocellulose membrane using semi dry transfer.
