Mouse Pms2 encodes the PMS2 protein, a core component of the MutLα mismatch repair complex with MLH1 that corrects base–base mismatches and insertion–deletion loops arising during DNA replication. PMS2 contributes endonuclease activity that coordinates excision and resynthesis steps, supporting genome stability and proper DNA damage signaling. Through its role in post-replicative repair and maintenance of microsatellite integrity, altered PMS2 function is linked to elevated mutation rates and dysregulated responses to replication stress. In research settings, Pms2 perturbation is used to model mismatch repair deficiency and to study mutation accumulation, DNA repair pathway crosstalk, and checkpoint responses.
PMS2 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Pms2 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Pms2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Pms2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish PMS2 protein expression.
This CRISPR knockout system enables efficient generation of Pms2-deficient cell models for investigation of PMS2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
