    Overview

    MUC6 encodes mucin 6, a secreted, heavily O-glycosylated gel-forming mucin that contributes to the protective mucus barrier on epithelial surfaces. Through its extensive glycan structures and polymeric matrix formation, MUC6 helps regulate hydration, viscosity, and interactions with microbes and luminal factors, shaping mucosal homeostasis and innate defense. Its expression is linked to epithelial differentiation and mucin biosynthesis programs that interface with ER/Golgi glycosylation pathways and barrier maintenance processes. Dysregulated MUC6 expression and altered mucin glycosylation patterns are associated with inflammatory injury and epithelial remodeling in gastrointestinal and other mucosa-associated diseases, and are frequently evaluated as biomarkers of lineage and mucous cell states in cancer biology.

    Mucin 6/MUC6 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the MUC6 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the MUC6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the MUC6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Mucin 6/MUC6 protein expression.

    This CRISPR knockout system enables efficient generation of MUC6-deficient cell models for investigation of Mucin 6/MUC6 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
