    Overview

    MUC5B encodes mucin 5B, a high–molecular weight, gel-forming secreted mucin that is a major structural component of airway and salivary mucus. Through extensive O-glycosylation and polymerization, MUC5B contributes to mucociliary clearance, pathogen trapping, and epithelial barrier homeostasis, and its expression is regulated by inflammatory and differentiation programs in secretory epithelia. Altered MUC5B abundance or rheological properties are linked to dysregulated mucus transport and chronic airway remodeling, and genetic and expression changes have been associated with lung fibrosis and muco-obstructive lung disease phenotypes. As a readout-rich secreted glycoprotein, MUC5B is widely studied in epithelial differentiation, ER/Golgi secretory processing, and innate defense signaling contexts.

    Mucin 5B/MUC5B CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the MUC5B gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the MUC5B together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the MUC5B open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Mucin 5B/MUC5B protein expression.

    This CRISPR knockout system enables efficient generation of MUC5B-deficient cell models for investigation of Mucin 5B/MUC5B signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
