IRF-1 CRISPR/Cas9 KO Plasmid (r) | sc-437365 | 20 µg | $397.00 |
Interferon regulatory factor 1 (IRF-1) is a DNA-binding transcription factor that integrates interferon, cytokine, and pathogen-sensing signals to shape innate and adaptive immune gene programs in rat cells. IRF-1 regulates transcriptional networks controlling antigen presentation, inflammatory mediator production, and antiviral responses, interfacing with JAK/STAT signaling and cross-talk with NF-κB-dependent pathways. Through its roles in cell cycle control and apoptosis, IRF-1 influences stress-response outcomes and immune cell differentiation. Altered IRF-1 activity has been associated with dysregulated inflammation and host defense phenotypes, supporting its use as a mechanistic node in immunology and infection models.
IRF-1 CRISPR/Cas9 KO Plasmid (r) is a pool of plasmids designed for targeted disruption of the gene in rat cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish IRF-1 protein expression.
This CRISPR knockout system enables efficient generation of -deficient cell models for investigation of IRF-1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
