O Anticorpo de Insulina B (C-12) é um anticorpo monoclonal de camundongo IgG1 κ para insulina B (também designado anticorpo de peptídeo de insulina, anticorpo de hormônio de crescimento de insulina) que detecta a proteína insulina B de origem de camundongo, rato e humana por WB, IP, IF, IHC(P) e ELISA. O Anticorpo Insulina B (C-12) está disponível na forma de anticorpo anti-Insulina B não conjugado, bem como em várias formas conjugadas de anticorpo anti-Insulina B, incluindo agarose, HRP, PE, FITC e vários conjugados Alexa Fluor®. A insulina é um hormônio peptídico secretado que provoca efeitos metabólicos, como elevados níveis de captação de glicose e síntese de glicogênio, levando a uma diminuição da concentração de glicose no sangue. A insulina é formada inicialmente como uma molécula precursora, a pré-proinsulina, que é posteriormente dividida em pró-insulina e, finalmente, no hormônio insulina madura. A insulina madura consiste em 51 aminoácidos, contidos em uma cadeia A e uma cadeia B que são conectadas por duas pontes de dissulfeto. Ela eleva a permeabilidade celular a monossacarídeos, aminoácidos e ácidos graxos. A insulina é secretada pelo pâncreas em níveis basais na ausência de estímulos exógenos, com secreção elevada em resposta à glicose. A ação da insulina é efetuada pela ligação da insulina aos receptores da superfície celular na membrana da célula-alvo. Os defeitos da insulina são a causa da hiperproinsulinemia e do diabetes mellitus tipo II.

Can I use SC-377071 for sandwich ELISA?

Thank you for your question. This antibody, sc-377071 Insulin B (C-12), has not been tested for sandwich ELISA, but it is recommended and warrantied for direct or indirect ELISA.

I have contacted SCBT south east asia branch roughly 2 weeks a go to enquire about the 10 ug sample free of charge as stated in the product information Insulin B (C-12): sc-377071. However, there is no response. Can you help me regarding this matter?

The expected molecular weight of Insulin is 6-7 kDa. Why 12 kDa? It´s not clear. Thank you.

Thank you for your question. The predicted molecular weight of 12 kDa is for the precursor of Insulin B which is 110 amino acids in length.

How can Insulin B Antibody (C-12): sc-377071 be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody is currently available conjugated to phycoerythrin (sc-377071 PE), fluorescein (sc-377071 FITC), Alexa Fluor® 488 (sc-377071 AF488) or Alexa Fluor® 647 (sc-377071 AF647).

Wide-field diffuse amacrine cells in the monkey retina contain immunoreactive Cocaine- and Amphetamine-Regulated Transcript (CART). Peptides. 2016 Oct;84:22-35). The isolated retina of a baboon was fixed for 60 minutes in 4% paraformaldehyde. Tissue was incubated for 9 days in a 1:1,000 solution of primary antibody with detergent and a blocking agent at 4 °C, washed, then incubated in an affinity-purified secondary antibody for 4 days at 4 °C, washed, and mounted in medium containing DAPI. No peptide blocking control was performed. Results: Very light punctate labeling and low background throughout the optic fiber layer, ganglion cell layer, inner plexiform layer, and inner nuclear layer (less).
