    H2-I-A/I-E Antibody (M5/114) is a rat monoclonal IgG2b antibody that detects H2-I-A/I-E protein of mouse origin by immunofluorescence (IF) and flow cytometry (FCM). Anti-H2-I-A/I-E antibody (M5/114) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. H2-I-A/I-E protein plays a crucial role in the immune system as a major histocompatibility complex (MHC) class II molecule, which is essential for presenting foreign peptides to cytotoxic T lymphocytes, thereby initiating an immune response. The structural characteristics of MHC class II molecules, including their ability to bind and present a diverse array of peptides, are vital for the activation of T lymphocytes, which are key players in adaptive immunity. M5/114 (M5/114.15.2) monoclonal antibody specifically reacts with mouse MHC class II glycoproteins encoded by the I-A and I-E subregions, including I-Ab, I-Ad, I-Aq, I-Ed, and I-Ek, while showing no reaction with I-Af, I-Ak, or I-As. Anti-H2-I-A/I-E antibody (M5/114) recognizes a polymorphic determinant present on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from mice with the H-2b, H-2d, H-2q, H-2p, H-2r, and H-2u haplotypes, but not from those with the H-2s or H-2f haplotypes. Additionally, H2-I-A/I-E monoclonal antibody (M5/114) has been reported to inhibit I-A-restricted T cell responses in mice with the H-2b, H-2d, H-2q, and H-2u haplotypes, highlighting its potential utility in immunological research and therapeutic applications.

    In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-23940 PE) or fluoroscein (sc-23940 FITC).
