E2F-1 Antibody (KH20) is a mouse monoclonal IgG2a kappa light chain antibody targeting E2F-1, also known as Retinoblastoma-associated protein 1 (RBAP-1) or Retinoblastoma-binding protein 3 (RBBP-3). Anti-E2F-1 antibody (KH20) is raised against a recombinant protein corresponding to amino acids 1–89 of human E2F-1 and is reactive with E2F-1 from human, mouse, and rat origins in western blotting (WB) and immunoprecipitation (IP) applications. E2F-1 is a pivotal transcription factor within the E2F family, playing a critical role in regulating genes essential for cell cycle progression, particularly the transition from the G1 to S phase, as well as DNA replication and apoptosis. Structurally, E2F-1 contains several conserved domains, including a DNA-binding domain, a dimerization domain for interaction with DP proteins, a transactivation domain rich in acidic amino acids, and a tumor suppressor protein association domain embedded within the transactivation domain. These structural features enable E2F-1 to form heterodimers with DP proteins, creating an active transcriptional regulatory complex that binds to specific DNA sequences to modulate gene expression. The interaction between E2F-1 and the retinoblastoma protein (pRB) is crucial for controlling cell proliferation, as pRB binding inhibits E2F-1 activity, preventing uncontrolled cell division. Dysregulation of E2F-1 has been implicated in various cancers due to its influence on cell cycle control and apoptosis.
