Polb encodes DNA polymerase β, a central enzyme in the base excision repair (BER) pathway that fills short DNA gaps after removal of damaged bases and processes 5′-deoxyribose phosphate intermediates. By coordinating with XRCC1, DNA ligase III, and other BER factors, DNA polymerase β helps maintain genome stability and limits accumulation of oxidative and alkylation-induced lesions. Disruption or dysregulation of BER components, including polymerase β, is associated with elevated mutational burden, impaired DNA damage signaling, and altered cellular stress responses. These functions make Polb a relevant target for studying DNA repair capacity, genotoxic sensitivity, and mechanisms linking repair defects to disease-associated genomic instability.
DNA pol β CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Polb gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Polb together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Polb open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish DNA pol β protein expression.
This CRISPR knockout system enables efficient generation of Polb-deficient cell models for investigation of DNA pol β signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
