KLRB1 encodes CD161 (KLRB1), a C-type lectin-like receptor prominently expressed on subsets of NK cells and T cells, including mucosal-associated invariant T (MAIT) cells and Th17-like populations. CD161 participates in immunoregulatory signaling and modulation of cytotoxicity and cytokine output through receptor-mediated pathways that shape innate and adaptive immune responses. Altered KLRB1/CD161 expression is associated with immune activation states and inflammatory contexts, and it is frequently used to phenotype functionally distinct lymphocyte subsets in studies of infection, autoimmunity, and tumor immunology. As a surface receptor linked to effector programs such as IL-17–skewed responses and tissue homing, CD161 provides a tractable entry point for dissecting lymphocyte differentiation and function.
CD161 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the KLRB1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the KLRB1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the KLRB1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish CD161 protein expression.
This CRISPR knockout system enables efficient generation of KLRB1-deficient cell models for investigation of CD161 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
