O anticorpo 8-oxoG DNA Lesion (483.15) é um anticorpo monoclonal 8-oxoG DNA Lesion IgM κ de camundongo (também designado anticorpo 8-oxoG DNA Lesion) que detecta a proteína 8-oxoG DNA Lesion. O anticorpo 8-oxoG DNA Lesion (483.15) está disponível como anticorpo não conjugado anti-8-oxoG DNA Lesion. O ADN (ácido desoxirribonucleico) é o material hereditário de todos os organismos vivos conhecidos e de alguns vírus. Essencial para o armazenamento de informação a longo prazo, bem como para o desenvolvimento e funcionamento, o ADN é constituído por dois polímeros de nucleótidos longos e antiparalelos que contêm espinhas dorsais de açúcar e grupos fosfato que são mantidos juntos por ligações éster, formando uma dupla hélice. A 8-oxoG (8-oxoguanina) é uma lesão mutagénica envolvida em danos oxidativos no ADN. A 8-oxoG é capaz de se emparelhar com a adenina (A) durante a replicação do ADN, constituindo uma ameaça para a estabilidade do genoma. Todos os organismos expressam um mínimo de dois tipos de 8-oxoG-DNA glicosilase (OGG) para reparar a 8-oxoG.

What is the recommended protocol for IF with this antibody? I tried methanol fixing (your standard guidelines) and IF with 1:50 dilution and standard secondary antibody, unfortunately didn' get any signal.

I want to use this antibody to detect 8-oxoG level on extracted mitochondrial DNA from c.elegans by performing dot blot. I'm wondering whether this antibody preferentially react to single stranded DNA and RNA. Do I need to denature my DNA sample?

Thank you for your question. This is an antibody against the DNA lesion only as it will detect the 8-oxoG lesion itself. We recommend this antibody for ELISA and IF, not Western blotting, and no denaturing should be necessary.

130914 8-oxoG is a IgM (Kappa light chain) why your company recommend use second ab is Anti mIgGk FITC or PE (516140 or 51614)? Do you have Anti mIGM k Ab? Can I you Anti mIGM ab as a second Ab?

Thank you for your question. We recommend the mouse IgG binding proteins for most IgM antibodies as they are fine for this use. If you have an anti-IgM secondary antibody, that will be fine to use as well.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with 8-oxoG DNA Lesion Antibody (483.15): sc-130914?

Rated 1 out of 5 by from Inconsistent from lot to lotI purchased a tube of anti-8-oxoG sc-130914 and optimized an immunofluorescence protocol using it in cultured human fibroblasts. The stain looked very good, and the signal was inducible by treatment with H2O2 or KBrO3 and repressible by treatment with Fpg. I used the whole tube for experiments and purchased a new tube, but the new tube performs terribly. It binds to all DNA so non-specifically that the signal is too strong to quantify even when the antibody is used at a dilution of 1 to 10,000 (I was using the first tube at 1 to 50). You might get lucky and get a good lot, but maybe you won't.

Rated 1 out of 5 by from No signal in IFI tested this antibody (purchased <2 months ago) on the cells subjected to 5mM, 1mM and 0 mM (negative control) H2O2 for 1 h twice: (1) using the standard methanol fixing procedure (without permeabilisaition) with buffers containing Triton X-100 0.3%; and (2) following the recommendation of SC Tech Support (https://www.scbt.com/de/resources/protocols/immunofluorescence-cell-staining) with the use of BSA as a blocking agent (the host of my secondary antibody routinely used in the lab is rabbit). In both experiments there's no signal above the 'no antibody' control. I am not an expert in this field but I have read that DNA denaturation is one of the steps of 5-mC IF procedure. Isn't it necessary also for detecting 8-oxoG?
