

TOX Antibody (G-5) is a mouse monoclonal IgG1 antibody that detects TOX in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TOX, or Thymocyte selection-associated high mobility group (HMG) box protein, is a crucial 526 amino acid nuclear protein belonging to the HMG box family of DNA-binding proteins, which are integral to the regulation of T-cell development. TOX (G-5) antibody recognizes proteins involved in modulating thymocyte transition through various developmental stages, particularly the critical shift from double-positive to single-positive T cells, which is essential for generating class II major histocompatibility complex-specific T cells. TOX expression increases with pre-T cell receptor and T cell receptor activation in immature thymocytes, highlighting a role in early T cell development. In TOX-deficient mice, the absence of CD4 T cell development underscores TOX′s importance in T cell lineage commitment, while functional CD8(+) T cells suggest TOX is specifically required for CD4(+) T cell maturation. Additionally, calcineurin activation links to TOX expression, as shown by calcineurin-dependent upregulation of TOX in double-positive thymocytes, indicating a complex interplay between signaling pathways governing T cell differentiation.
